Senti Bio Receives CIRM Grant to Advance Logic Gated CAR-NK Cell Therapy in Clinical Development

Senti Biosciences, Inc. announced today that it has received an $8 million grant from the California Institute for Regenerative Medicine to advance its Phase 1 clinical trial of SENTI-202. This innovative therapy, designed for the treatment of relapsed/refractory hematologic malignancies such as acute myeloid leukemia (AML), utilizes a Logic Gated CAR-NK cell approach.

SENTI-202 targets CD33 and/or FLT3 expressing cancer cells while preserving healthy bone marrow cells. It incorporates an activating CAR (OR GATE) to eliminate leukemic cells, a protective mechanism (NOT GATE) to spare healthy cells, and calibrated-release IL-15 technology to enhance cell persistence and activity. The therapy uses NK cells sourced from screened adult donors and cryopreserved for consistency in manufacturing.

The Phase 1 trial (NCT06325748), currently ongoing in the United States and Australia, is evaluating two dose levels of SENTI-202 administered in cycles after lymphodepleting conditioning. Initial efficacy data is expected by late 2024, with durability data anticipated in 2025.

Acute myeloid leukemia (AML) is the most common type of acute leukemia in adults, with an estimated 20,800 new cases expected in the U.S. in 2024. Current treatment options for relapsed or refractory AML are limited, highlighting the potential significance of SENTI-202 as a new therapeutic approach.
