Logopharm provides specialized services in proteomic target and biomarker R&D with a strong focus on membrane proteins, protein complexes and antibody validation. Founded in 2005 as a spinout of the University of Freiburg, Germany, the company combines most advanced technology and instrumentation with long-term experience in membrane protein analysis and molecular physiology to tackle even the most challenging targets, like ion channels, G-protein coupled receptors or other membrane receptors and transporters. Proteomic approach

The underlying proteomic approach is distinct from current high-throughput technologies that are based on recombinant systems and therefore suffer from high error rates, in that

  • Logopharm uses native tissues and cells as source material to isolate protein isoforms and assemblies forming under physiological conditions,
  • The biochemical tools and protocols are highly optimized and have been validated in multiple target projects, and
  • Proteins are analyzed in an unbiased manner by quantitative high-resolution mass spectrometry (LC-MS/MS) with ultimate sensitivity

Micro-affinity purification

High Content Antibody Profiling

Even with careful design of epitopes and selection techniques it isimpossible to predict the in-vivo binding properties of antibodies towardsnatively folded proteins. High content antibody profiling through Logopharm's proprietary parallel multi-affinitypurification (PMAP) platform can determine the relative off-targetreactivity, isoform selectivity and target binding efficiency of up to 96antibodies/experiment in a native-like environment. This provides a rational basis for antibody selection and epitope design.

Protein complexes

Protein-protein interactions and signaling pathways

The CompleXio platform has been specifically designed for identification of protein-protein interactions in native tissues and cells. It is based on nativesolubilisation of protein complexes followed by affinity-purification with multiple antibodies and stringent controls (preferably including target knockouts), identification by high-performance mass spectrometry (LC-MS/MS) and quantitative evaluation of results.

ComplexioLytesolubilisation buffers

ComplexioLytesolubilisation buffers from Logopharm are ready-to-use detergent buffers for efficient solubilisation of membrane proteins and protein complexes. These buffers have been successfully applied in affinity purifications, native gel electrophoresis, size exclusion chromatography and (non-)denaturing FFE.ComplexioLyte testing assays include:

  • Ultracentrifugation separation and western blot analysis for determining relative solubilisation efficiency.
  • 2D gel electrophoretic separation (native PAGE/SDS-PAGE) of solubilisates revealing target complex size distribution and integrity of reference complexes.
  • Affinity purification coupled with LC-MS/MS analysis to confirm preservation of known associated partner proteins.

Membrane protein tools

Microproteomicsanalysis

Microproteomics analysis provides ultimate proteomic sensitivity (lowfemtomolar range) and efficiency (down to some 100.000 cells). This platform comprises miniaturized biochemistry, optimized protocols and tools as well as new technologies (parallel micro affinity purification, PMAP) and highly sensitive mass spectrometry.It has beensuccessfully employed to study membrane proteins in systems previously notaccessible to functional proteomics, such as dorsal root ganglion cells, thecochlear Organ of Corti or hippocampal slices collected by laser-capturemicrodissection.

Proprietary protein target database and track record

Customers and partners may also benefit from Logopharm's large internal protein database. This includes more that 2000fully MS-analyzed affinity purifications of membrane proteins and theirinteractors, covering a major portion of the expected mammalian membrane proteome. Thus, it is a valuable resource for technical, biochemical as well as target-related information, such as marker peptides, solubility, abundance/variation, tissue specificity, interaction partners, physiological context and modifications.

Logopharm strategy

Annotation of uncharacterized / "orphan" proteins

About a third of the mammaliangenome codes for functionally unassigned proteins, and another third has been annotated based only on homology predictions. The unbiased identification of protein partners by Logopharm's approach can offer a direct access to study function of these proteins in their physiological context and to establish screening assays.

Protein trace analysis

Logopharm's mass spectrometric quantification methodology allows for sensitive quantification of proteins over a range of at least four orders of magnitude (0.1-1000 fmoles). This makes protein trace analysis especially useful for identification and quantification of protein contaminations in biopharmaceuticals, in orderto improve the production process and exclude critical contaminations.
